Roger Federer was the two-time defending champion, but lost in the semifinals to Novak Djokovic. Djokovic won in the final 4–6, 6–4, 6–3, 7–6(7–2), against an unseeded Jo-Wilfried Tsonga (ranked no. 38) playing in his first ATP final. It was the first Grand Slam title of Djokovic's career, and the first of his eventual six Australian Open titles. It was also the first Grand Slam final since the 2005 Australian Open that did not feature either Federer or Rafael Nadal, who were beaten by Djokovic and Tsonga, respectively, in the semi-finals. This ended Federer's run of a record 10 straight Grand Slam finals. He would go on to make the next 8 Grand Slam finals, for a run of 18 Grand Slam finals in a span of 19 tournaments.

Serena Williams was the defending champion, but she was defeated in the quarterfinals by Jelena Janković. Maria Sharapova, the runner-up to Williams in 2007, won in the final 7–5, 6–3, against Ana Ivanovic to win her third Grand Slam title and her first at the Australian Open. She was also the first Russian woman to win this tournament. Ivanovic lost in a Grand Slam final for the second time, though she would eventually win the French Open a few months later.

Rafael Nadal was the 3-time defending champion and retained his title, defeating Roger Federer in the final 6–1, 6–3, 6–0. This was Federer's worst loss in a Grand Slam event in his entire career. It also marked the third consecutive year that Nadal defeated Federer in the Roland Garros final, and the fourth consecutive year that Nadal defeated Federer at Roland Garros, starting with his 2005 Roland Garros semifinal defeat. All four years, Rafa defeated Federer while he was the No. 1 player in the world. Nadal won the title without losing a set, becoming the third man in the Open era after Ilie Năstase and Björn Borg to win the French Open without dropping a set for the entire tournament. He would duplicate the feat in 2010 and 2017 at Roland Garros, as well. This was the final Grand Slam and World Tour appearance of former world No. 1 and three-time champion Gustavo Kuerten. He lost to Paul-Henri Mathieu in the first round and retired from professional tennis shortly after.

Justine Henin was the three-time defending champion, but she retired from the sport on May 14, 2008. Her retirement left Serena Williams as the only former French Open champion remaining in the women's draw. Williams was defeated in the third round by Katarina Srebotnik, meaning that there would be a first-time French Open champion in 2008. Ana Ivanovic, the runner-up to Henin in 2007, won her first and only Grand Slam singles title, beating first-time finalist Dinara Safina, 6–4, 6–3 in the final. Ivanovic went through the entire tournament dropping only one set, against Jelena Janković, in an all-Serbian semifinal which decided the world No. 1 ranking. Ivanovic subsequently displaced Maria Sharapova from the top ranking, who had been elevated to world No. 1 ahead of the tournament following Henin's retirement. Safina had come from a set and match point down in both her fourth round and quarterfinal matches against Sharapova and Elena Dementieva, respectively. This was also the first Grand Slam main draw appearance of future World No. 2 and two-time grand slam champion Petra Kvitová.

In the 2008 Wimbledon Championships Men's Singles Final, Rafael Nadal defeated five-time defending Wimbledon champion, Roger Federer in five sets to win his first Wimbledon title and fifth Grand Slam championship. It was the longest championship match in Wimbledon history with play lasting four hours and 48 minutes, and with two rain delays. The event stretched over seven hours ending just before nightfall, and is widely considered to be the greatest match in tennis history.

Venus Williams was the defending champion, and won in the final 7–5, 6–4, against her sister Serena Williams. This was Venus's fifth Wimbledon title and her seventh Grand Slam singles title overall. This was also the second time she won a Grand Slam tournament without losing a set. Zheng Jie became the first Chinese player of either sex to reach a Grand Slam singles semifinal, having defeated the world No. 1 Ana Ivanovic en-route in the third round. With the other top four seeds—Jelena Janković, Maria Sharapova and Svetlana Kuznetsova—also losing in the first four rounds, it marked the first time in Wimbledon history that none of the top four women's seeds had advanced to at least the quarterfinals.

Roger Federer was the four-time defending champion. He won a 5th consecutive US Open title after winning the final 6–2, 7–5, 6–2 against Andy Murray of Great Britain who was contesting his first major final. It was Federer's 13th major title and his only successful title defence in majors that year, after losing the Australian Open and Wimbledon titles. Rafael Nadal had a chance to win Grand Slams on clay, grass and hard court in the same calendar year, after having won on this year's French Open and as well as Wimbledon and could become the first man to win French Open, Wimbledon and US Open since Rod Laver did in 1969, but he lost to Murray in the semifinals. Future champion Marin Čilić appeared in the main draw of the US Open for the first time.
